Abstract This manuscript deals with a hierarchical control problem for Oldroyd equation under the Stackelberg-Nash strategy. The Oldroyd equation model is defined by non-regular coefficients, that is, they are bounded measurable functions. We assume that we can act in the dynamic of the system by a hierarchy of controls, where one main control (the leader) and several additional secondary control (the followers) act in order to accomplish their given tasks: controllability for the leader and optimization for followers. We obtain the existence and uniqueness of Nash equilibrium and its characterization, the approximate controllability with respect to the leader control, and the optimality system for leader control.

Biological evaluation of benzothiazoles obtained by microwave-green synthesis OZDINCER, MESUT DALMAZ, ASLIHAN DURMUS, SEFA DULGER, GORKEM KILICCIOGLU, ILKER

Abstract Benzothiazole compounds are known as an important bicyclic ring system with multiple applications. These compounds have a wide range of biological activities, including anticancer, antimicrobial, anti-inflammatory and antiviral activities. In this study, benzothiazole compounds were synthesized and their various biological activities were examined. The synthesized benzothiazoles were evaluated for their antimicrobial properties against various bacterial and fungal strains. The compound 6e is most active ligand in the series against bacteria and fungi as compared to standard antibiotics. Especially, this compound significant effect against Staphylococcus aureus (32.00 ± 1.73 mm). These compounds exhibited potent anticancer activity against gastrointestinal cancer cells, demonstrating their potential as therapeutic agents. The lowest antiproliferative response after administration of the compounds was observed in HCT116 cells, while the most effective antiproliferative response was observed in AGS cells (> 10 µg/mL). In all cell lines, 40 and 100 µg/mL application values of the selected compounds showed significant increases in the expression of caspase-3, 8 and 9. We also utilized a computational docking approach to investigate the interaction of these benzothiazoles with VEGFR-2 kinase. Our docking studies showed that compounds 6a and 6d may be promising therapeutic agents against gastrointestinal system cancers due to their ability to bind to VEGFR-2 kinase.

Abstract Fermentation using starter cultures has been considered an alternative and economically viable technology for the production of specialty coffees. This type of technology promotes several benefits, such as increased sensory quality, control over the fermentation process, predictability of the final product and added value. Coffee (Coffea arabica L.) samples for this study were collected in Presidente Olegário - MG (2018/19 crop year) in the Cerrado region of Minas Gerais. The effects of natural fermentation and inoculation of the yeast Torulaspora delbrueckii and duration of fermentation (0, 24, 48, 72 and 96 hours) on the sensory and chemical quality (analysis of bioactive, volatile, and organic compounds and fatty acids) of coffee were evaluated. The objective of this study was to determine the effect of fermentation time and starter culture inoculation on the chemical composition of fermented coffees. Fermentation time significantly influenced the sensory description of the coffee beverage, with notes of honey, brown sugar and almond predominating up to 48 hours, for coffees fermented for 72 and 96 hours the notes described were and fruity, winey notes. The chemical composition was primarily influenced by fermentation time.

Analysis of Tribology Properties of Trimethylolpropane-based Lubricant by Molecular Dynamics SOUZA, CAMILA B. GONÇALVES, RENE FRANCISCO B. ROCCO, JOSÉ ATÍLIO F.F.

Abstract Currently, it is crucial for the lubricant formulation industry to explore cost-effective and environmentally friendly methodologies for analyzing the tribological properties of engine aviation lubricants under high-temperature and high-pressure operating conditions. This study demonstrates the feasibility of employing molecular dynamic simulations to gain essential insights into the evolution of the tribological properties of lubricants during operation. A three-layer molecular model was devised, comprising nickel aluminide molecules in the top and bottom layers, and polyol ester in the core. The impact of sliding velocities ranging from 20 km/h to 100 km/h was investigated under varying temperature and pressure conditions. Concentration, temperature and velocity profiles, radial distribution function, mean square displacement, and friction coefficient were calculated and analyzed in detail. Notably, the highest friction coefficients – ranging from 2.5 to 0.75 - were observed at the lowest temperature and pressure conditions tested. Conversely, other sections of the gas turbine exhibited substantially lower friction coefficients – ranging from 0 to 0.01.Simulations demonstrate that increasing pressure and temperature reduce polymer chain mobility, leading to stronger internal interactions within the lubricant. Consequently, lubricant adsorption onto metal surfaces decreases. Furthermore, the lubricant performs exceptionally well when its molecules encounter higher velocities and temperatures. Based on the results obtained, the research demonstrates that the presented technique provides both quantitative and qualitative tribological information essential for understanding a system molecular behavior, serving as a guiding framework for researchers in the field.

Abstract Jacobsite is a relatively rare mineral of composition MnFe2O4, found in Urandi (Bahia State) in Brazil. It is also a common species in the deep-sea manganese nodules, attracting the interest of many mineral-extracting companies. Because of its spinel constitution similar to magnetite, Jacobsite is commonly called a manganese-ferrite. However, the manganese/iron content may vary substantially according to its origin, demanding specific studies in each case. The Jacobsite mineral inspired our laboratory synthesis of the analogous manganese ferrite nanoparticles. The direct synthesis by the coprecipitation method has not been successful; however, it can be carried in the presence of citrate ions, yielding strongly magnetic nanoparticles, with a maximum magnetization of 45.6 emu.g1. Although they were structurally identical to Jacobsite, the mineral from Bahia exhibited a rather weak magnetism, because it involves a ferrimagnetic coupling. For this reason, the synthetic method seems to provide a better way of obtaining strongly magnetic manganese ferrites. These magnetic nanoparticles have been investigated in detail, including their interaction with diatoms, providing interesting magnetic bio-silicate carriers in drug delivery.

Abstract This study aimed to investigate the antioxidant and anti-inflammatory properties of quercetin on the cellular components of the Enteric Nervous System in the ileum of rats with arthritis. Rats were distributed into five groups: control (C), arthritic (AIA), arthritic treated with ibuprofen (AI), arthritic treated with quercetin (AQ) and arthritic treated with both ibuprofen and quercetin (AIQ). The ileum was processed for immunohistochemical techniques for HuC/D, calcitonin gene-related peptide, and vasoactive intestinal polypeptide. Measurements in histological sections, chemiluminescence assays, and total antioxidant capacity were also performed. Rheumatoid arthritis resulted in a decrease in neuronal density, yet neuroplasticity mechanisms were evident through observed changes in varicosities size and neuronal area compared to the control group. Reduced paw edema and neuroprotective effects were predominantly noted in both plexuses, as evidenced by the increased density preservation of HuC/D-IR neurons in the AIQ group. The increase of lipoperoxidation levels and paw edema volume in the AQ group was observed compared to the arthritic, whereas the AIQ group mainly showed similar results to those observed in the control. The enteropathy associated with arthritis proved to be significant in the field of gastroenterology, and the combination of quercetin and ibuprofen demonstrated promising anti-inflammatory and neuroprotective effects.

Abstract Data about the impacts of hemodialysis on antioxidant status and markers of oxidative stress are controversial, probably due to the use of different methodological approaches. The aim of this study was to assess the changes in the oxidative damage markers and antioxidant enzymes, and the serum antioxidant capacity by using in vitro model systems of free radical generation before and after one hemodialysis session. Blood samples were collected from 40 patients with kidney failure before and after hemodialysis. In pre- and post-hemodialysis serum samples, concentrations of biomarkers of oxidative damage and the activities of antioxidant enzymes were measured, as well as the in vitro antioxidant potential. The high concentrations of oxidative stress markers in serum of kidney failure patients were decreased after one hemodialysis session. In pre-hemodialysis, low activities of antioxidant enzymes were observed, including paraoxonase-1, however paraoxonase-1 activity was partially recovered after hemodialysis. Crocin bleaching and radical scavenging assays showed that serum antioxidant potential was decreased after hemodialysis. Although one hemodialysis session increased paraoxonase-1 activity and decreased oxidative stress markers, it caused a decrease in the serum antioxidant potential. Future research is needed to prospect strategies to mitigate the impacts of oxidative stress in the scenario of hemodialysis repetitions.

Abstract Nutritional insults early in life, such as during the suckling phase, are associated with phenotypic alterations and promote adverse permanent effects that impair the capacity to maintain energy balance in adulthood. This study aimed to evaluate the long-term effects of a low-protein (LP) diet during lactation on the metabolism and antioxidant systems of adult female rat offspring. Dams were fed a low-protein diet (4% protein) during the first two weeks of lactation or a normal-protein (NP) diet (20% protein) during the entire lactation period. The female offspring received a standard diet throughout the experiment. At 90 days of age, female LP offspring exhibited decreased body weight, feeding efficiency, and fat pad stores. The adult LP female offspring displayed brown adipose tissue hyperplasia without alterations in glucose homeostasis. The LP diet decreased liver triglyceride content and improved the antioxidant system compared to the NP group. The LP diet during the suckling phase promotes a lean phenotype and improves the hepatocyte antioxidant system in adult female offspring. Thus, the LP diet may play an important role in homeostasis and the prevention of metabolic damage.

Phosphokinases related to drug resistance in two cohorts from the cancer genome atlas (TCGA): uterine carcinoma and testicular cancer OLIVEIRA, BRUNO R. MARQUES, MAIARA B. WERHLI, ADRIANO V. MARINS, LUIS FERNANDO

Abstract We aimed to find new therapeutic targets related to Cancer Stem Cell alterations in recurrent patients from two TCGA cohorts: Testicular Germ Cell Tumor (TGCT) and Uterine Corpus Endometrial Carcinoma (UCEC). Raw sequencing data were downloaded from the TCGA database. Datasets containing RNA expression and Methylation files were directly downloaded from cBioportal. Variant Call Format files (VCFs) were downloaded from the GDC portal. Gene enrichment analysis was performed using GSEA (Gene Set Enrichment Analysis) software. Transcriptome profiling, coexpression co-occurrence, networks, and survival analyses were performed using cBioportal tools, while mutational analysis of patients was processed using UNIX scripts. We found that cancer stem cell transcription factors were highly expressed in Testicular Germ Cell Tumor (TGCT) and Uterine Corpus Endometrial Carcinoma (UCEC) cohorts, compared to the other 29 cancer cohorts in TCGA. Patients presented a poorer diagnosis when the genes (POU5F1, NANOG, SOX2, SALL4, ABCB1, ABCC1, and ABCG2) were altered. In UCEC cohorts, recurrent patients showed the ABCG2 potentially phosphorylated by the PIM1 kinase. In the TGCT cohort, genes ABCB1 and ABCG2 only appeared in the phosphonetwork in recurrent patients potentially phosphorylated by the same kinase, PIM1, but also by PRKACA. Our data indicate that PRKACA and PIM1 may modulate POU5F1 phosphorylation.

Abstract It was compared smartphone-based measurements of static balance control and mobility of elderly population with and without type 2 diabetes mellitus (DM2). The present cross-sectional study investigated 73 participants grouped in a control group (n = 36) and a DM2 group (n = 37). Smartphone’s built in inertial sensors were used to record inertial changes of the participants during static balance and mobility (Timed Up and Go test – TUG) tasks. The inertial variations as a function of the time were analyzed and compared between groups. Both groups were matched in age, body mass index, male-female proportion, but DM2 group had significant larger fasting glucose than control group. Additionally, DM2 group had worst static balance control with open and closed eyes than the controls (p < 0.05) as well as they also had longer duration to execute the different events of the mobility test than the controls (p < 0.05). DM2 patients had decline of motor functions compared to controls and the use of bult-in sensors of smartphones was feasible to identify these functional impairments. The easy access of smartphones could be improving the screening of functional impairments in DM2 patients.

Insights into the association of H1N1 seasonality with the COVID-19 pandemic in Brazil: an ecological time series analysis SIMEONE, DIEGO GUIMARÃES-COSTA, AURYCÉIA

Abstract During the COVID-19 pandemic, H1N1 seasonality disappeared worldwide. In Brazil, information on how coronavirus impacted this seasonality is scarce. In this study, we aimed to verify whether COVID-19 pandemic was associated with changes in the seasonality of H1N1, modeling the time series of H1N1 between pre-pandemic (2018 and 2019), pandemic (2020 and 2021) and post-pandemic (2022 and 2023) periods. For this purpose, we superimposed on this time series cases of COVID-19 from 2020 to 2023. Our findings highlighted that H1N1 exhibited a consistent seasonal pattern in the pre-pandemic period, with peaks mainly in months with the highest rainfall. However, this seasonality disappeared during the pandemic, with a significant decrease in the number of cases, in contrast with the predicted seasonality of H1N1 for the same period. In addition, the seasonal pattern of H1N1 in the post-pandemic showed a return to that observed in the pre-pandemic period, especially in 2023. We observed that the COVID-19 pandemic was consistently associated with changes in H1N1 seasonality in Brazil, underscoring the relative importance of monitoring patterns of respiratory syndromes to enhance our understanding of how coronavirus is associated with changes in seasonal diseases.

Caffeine and Ovariectomy Debilitate Bone Health in Rats on Calcium-deficient Diet FRANCO, FREDERICO S.C. NATALI, ANTÔNIO JOSÉ COSTA, NEUZA MARIA B.

Abstract This study investigated the effects of ovariectomy and caffeine intake on bone health in rats on calcium-deficient diet. Forty adults female Wistar rats were divided into 4 groups in a 2x2 factorial design: Ovary (OVX/SHAM) and Caffeine (placebo/caffeine). The animals were housed in individual cages for 8 weeks, receiving 18-20g of AIN-93M diet per day, containing 50% of the daily recommended intake of calcium. The rats underwent ovariectomy (OVX) or laparotomy (SHAM) surgery. Caffeine groups received 6mg of caffeine/kg/day. After euthanasia, the tibia and femur were dissected to determine the calcium content and bone fracture strength, respectively. Blood sample was collected to determine serum Ostase. 24-hour urine was analyzed for excreted calcium and NTx. Reduced bone fracture strength and calcium content were observed in OVX and Caffeine groups. When observed separately, OVX group showed increased urinary NTx and lower bone weight, blood ostase, and urinary calcium. Caffeine groups showed elevated urinary calcium. There was a positive correlation between bone fracture strength and calcium content. NTx correlated negatively with bone calcium, fracture strength and thickness. In conclusion, both OVX and caffeine intake debilitate bone health in rats on calcium-deficient diet.

Photobiomodulation and amniotic membrane for treat tendon injury in rats NICODEMO, MARIANA C. ARISAWA, EMILIA ANGELA L.S. SANT’ANNA, LUCIANA B. LOPES-MARTINS, RODRIGO

Abstract Tendons, complex fibrous structures, are subjected to great tensions, which can give rise to the so-called tendinopathies. This study aimed to evaluate photobiomodulation and human Amniotic Membrane applied as single or combined therapies to treat induced Achilles tendon lesions. Seventy-five rats were divided into five groups (n=15): C- control Sham surgery; I- tendon injury; LA- tendon injury treated with photobiomodulation; AM- tendon injury treated with Amniotic Membrane; LAM- tendon injury + photobiomodulation and Amniotic Membrane, subdivided into three groups (n=5) with analysis at 3, 7, and 14 days. The tendon injuries were made with a 20 g weight released from a mini guillotine onto the ankle in dorsiflexion. AM and LAM groups received an Amniotic Membrane fragment while LA and LAM groups received transcutaneous photobiomodulation, using a 660 nm wavelength laser. The inflammatory cells showed statistical differences between groups C and I (p<0.05), I and AM (p<0.01), I and LA (p<0.05), and I and LAM (p<0.01). Both photobiomodulation and Amniotic Membrane were shown to enhance tendon repair, and the association of photobiomodulation plus Amniotic Membrane was the most effective treatment. We conclude that the association of photobiomodulation plus Amniotic Membrane was effective in accelerating and improving the tendon regeneration process.

Abstract The use of platelet-rich plasma (PRP) and adipose-derived stromal cells (ADSC) have been investigated as a form of wound healing enhancement. The objective of this work was to evaluate the association of red propolis (RP) and PRP as inducers of ADSC for application in tissue regeneration. Adipose tissue post-collection and post-cryopreservation was isolated with type II collagenase, characterized by flow cytometry, and differentiated into osteogenic, chondrogenic and adipose cell. The viability of ADSC was evaluated when exposed to different concentrations of RP using the MTT and trypan blue assay. Acridine orange and ethidium bromide (AO/EB) was performed to evaluate cell death events. Horizontal migration methods were investigated in ADSC using autologous and homologous PRP associated with RP (PRP/RP). All assays were processed in triplicate. Flow cytometry and cellular differentiation showed that type II collagenase was effective for isolating ADSC post-collection and post-cryopreservation. RP extracts at concentrations of up to 50 μg.mL-1 presented no cytotoxic effects. Association of PRP and RP at 25 and 50 μg.ml-1 influenced ADSC migration, with total closure on the seventh day after exposition. The results here presented could stimulate proliferation of ADSC cells that may contribute directly or indirectly to the reconstructive process of tissue regeneration.

Abstract According to a biopsychosocial framework, personal and environmental factors might be mediators or facilitators/barriers, respectively, to functioning. However, it is not known how these factors can impact independence in household chores in children/adolescents with Down syndrome (DS). This study explored whether and how personal/environmental factors are associated with the independence level in household chores of children/adolescents with DS in Brazil. Caregivers of twenty-eight children/adolescents with DS were interviewed using the CHORES and a standardized questionnaire about personal (child’s age and sex) and environmental (socioeconomic level and maternal schooling) factors. Multiple linear regression analysis identified if/how these factors are associated with level of independence. For CHORES self-care and CHORES total, sex was a significant variable explaining 21.8% and 15.8%, respectively, of the variation in the outcomes. For the outcome CHORES family care none of the variables was significant. Female sex was associated with a lower need for assistance. We conclude that only the personal factor assessed related to female sex in children with DS was associated with the independence level in household chores. This finding highlights the importance of health care providers and families to encourage the independence in chores regardless of sex and promote opportunities for both boys and girls.

Potential developmental effects of licit and illicit substances in humans: An approach to risk-specific dose and incidence SANTOS JR, ARNALDO R. LIMA, CAROLLINE M.

Abstract Teratogens encompass any agent capable of causing a birth defect or elevating the incidence of defects within the population. This category includes substances like drugs, both legal and illegal. These substances cause congenital anomalies depending on the stage of development at the time of exposure, the dose, and the exposure time associated with the embryo. The most sensitive period is the embryonic stage, when the three leaflets give rise to tissues and organs. Susceptibility to teratogenesis decreases during the fetal phase but morphological and functional disturbance of the fetus may still occur. Substance use during pregnancy and its adverse effects are a public health problem and the lay population does not have access to this information. Particularly concerning is the period within the first six weeks of pregnancy, often before a woman realizes she is pregnant. Developmental data for many substances are simply not available, which makes the problem more serious. The aim of this study is to reflect on the teratogenic effects of licit and illicit substances in humans, focusing particularly on the dose that can induce malformations and their incidence in humans.

Abstract Plasmodium falciparum is known to cause severe malaria, current treatment consists in artemisinin-based combination therapy, but resistance can lead to treatment failure. Knowledge concerning P. falciparum essential proteins can be used for searching new antimalarials, among these a potential candidate is shikimate dehydrogenase (SDH), an enzyme part of the shikimate pathway which is responsible for producing endogenous aromatic amino acids. SDH from P. falciparum (PfSDH) is unexplored by the scientific community, therefore, this study aims to establish the first protocol for active PfSDH expression. Putative PfSDH nucleotide sequence was used to construct an optimized expression vector pET28a+PfSDH inserted in E. coli BL21(DE3). As a result, optimal expression conditions were acquired by varying IPTG and temperature through time. Western Blot analysis was applied to verify appropriate PfSDH expression, solubilization and purification started with lysis followed by two-steps IMAC purification. Enzyme activity was measured spectrophotometrically by NADPH oxidation, optimal PfSDH expression occur at 0.1 mM IPTG for 48 hours growing at 37 °C and shaking at 200 rpm, recombinant PfSDH obtained after purification was soluble, pure and its physiological catalysis was confirmed. Thus, this study describes the first protocol for heterologous expression of PfSDH in soluble and active form.

Abstract The genus Stylosanthes has economic importance in semi-arid regions and requires studies that reveal complex relationships between species involving different ploidies. The aim of this study was to cytogenetically investigate accessions of Stylosanthes identified as S. scabra, in order to properly identify the number, morphology, and pattern of distribution of heterochromatin, analyzing the karyological variability of these species. Accessions with 2n=40 and 2n=20 were identified, exhibiting semi-reticulate interphase nuclei, symmetric karyotype, varied morphology, with differences in average chromosomal size, and genome length. The analysis with the fluorochromes chromomycin (CMA) and 4’,6-diamidino-2-phenylindole (DAPI) allowed the visualization of two CMA+ blocks in the subterminal region of the short arm in all accessions, and DAPI+/CMA- bands in S. scabra accessions. This suggests that not only chromosomal rearrangements, but also changes in the composition of heterochromatin, may have occurred during the speciation of this genus, and that S. scabra may be undergoing chromosomal evolution based on the observed karyological differences. In addition to the ploidy level, the distribution pattern of CMA+ heterochromatin reinforces the separation between S. scabra and S. seabrana. Thus, this genus represents an interesting group of plants for further studies on the content and quantity of repetitive and non-repetitive DNA sequences.

Abstract Repetitive sequences can lead to variation in DNA quantity and composition among species. The Orchidaceae, the largest angiosperm family, is divided into five subfamilies, with Apostasioideae as the basal group and Orchidoideae and Epidendroideae showing high diversification rates. Despite their different evolutionary paths, some species in these groups have similar nuclear DNA content. This study focuses on one example to understand the dynamics of major repetitive DNAs in the nucleus. We used Next-Generation Sequencing (NGS) data from Apostasia wallichii (Apostasioideae) and Ludisia discolor (Orchidoideae) to identify and quantify the most abundant repeats. The repetitive fraction varied in abundance (27.5% in L. discolor and 60.6% in A. wallichii) and composition, with LTR retrotransposons of different lineages being the most abundant repeats in each species. Satellite DNAs showed varying organization and abundance. Despite the unbalanced ratio between single-copy and repetitive DNA sequences, the two species had the same genome size, possibly due to the elimination of non-essential genes. This phenomenon has been observed in other Apostasia and likely led to the proliferation of transposable elements in A. wallichii. Deep genome information in the future will aid in understanding the contraction/expansion of gene families and the evolution of sequences in these genomes.

Abstract Using Amazonian fruits to flavor kombuchas is a promising proposal, as it adds nutritional value to the drink. This work sought to develop kombucha flavored with Amazonian fruits and evaluate the bioactive compounds and antioxidant capacity of the formulations. Three kombucha formulations were prepared using green tea (Camellia sinensis) and three Amazonian fruits: cupuassu (Theobroma grandiflorum), tapereba (Spondias lutea L.) and bacuri (Platonia insignis). Kombucha fermentations were evaluated before and after the insertion of nectars through the analysis of phenolic compounds, vitamin C and antioxidant capacity. Analyzes of pH, total sugars, acetic acid, ethanol, and microbiological characterization of final formulations were also carried out. For the first fermentation, were found values of phenolic compounds and antioxidant capacity of 30.60 ± 0.93 mg EAG/L and 295.02 ± 5.59 µmol ET/mL, and the formulation with tapereba showed the highest values for total phenolic compounds (34.92 ± 12.25 mg EAG/L), antioxidant capacity (320.57 ± 9.53 µmol ET/mL) and vitamin C (198.25 mg/100g). Thus, the formulations developed had a crucial nutritional appeal to stimulate consumption by the population, in addition to enabling the valorization and addition of commercial value to the Amazonian fruits used.

Unusual plant-extract based media for the differentiation between species in the Candida albicans complex: A comparative study MORALES-LÓPEZ, SORAYA MACÍAS, LISAHIDY ELLES, DANNA YEPES, JAYR

Abstract Candida albicans is the most common agent in human fungal infections; nevertheless, in the last decades, the closely related yeasts Candida dubliniensis and Candida africana have emerged as pathogens. The purpose of this study was to compare tobacco agar with another five agars prepared from plant extracts (Origanum vulgare, Rosmarinus officinalis, Solanum rudepannum, Solanum oblongifolium and Brugmansia arborea) on the differentiation of C. albicans complex. The hyphae and chlamyconidia formation and the color and margin of the colonies of 200 clinical isolates of C. albicans, C. dubliniensis and C. africana were evaluated. After seven days of incubation at 28 °C, Tobacco agar, S. rudepannum and B. arborea agars allowed the differentiation of 100 % C. dubliniensis. Additionally, 24% of C. africana isolates produced brownish colonies in the medium prepared from Rosmarinus officinalis (rosemary) extract. These results indicate that S. rudepannun, B. arborea and rosemary agar could be used as screening for the phenotypic differentiation between the species of C. albicans complex. Rosemary agar could be used to aid in the differentiation of C. albicans from C. africana. These culture media based on plants, could be used as simple and inexpensive screening methods in the phenotypic differentiation of C. dubliniensis and C. africana.

Abstract Cardiovascular diseases, resulting from the deposition of clots in blood vessels, are the leading cause of death worldwide. Fibrinolytic enzymatic activity can catalyze blood clot degradation. Findings show that 36 fungal isolates recovered from Caatinga soils have the potential to produce fibrinolytic protease under submerged conditions. About 58 % of the isolates displayed fibrinolytic activity above 100 U/mL, with Mucor subtilissimus UCP 1262 being the most active. The protease was biochemically and biophysically characterized, showing that the enzyme had a high affinity for SAApNA substrate and was significantly inhibited by fluoride methyl phenyl sulfonyl-C7H7FO2S, suggesting that it is a chymotrypsin-like serine protease. The highest enzyme activity was detected at pH 5.0 and 28 °C. This fibrinolytic protease’s far-UV circular dichroism (CD) showed that its secondary structure was primarily α-helical. The purified fibrinolytic enzyme may represent a novel therapeutic agent for treating thrombosis. At temperatures above 65 °C, the enzyme lost all its secondary structure. Its melting temperature was 58.1 °C, the denaturation enthalpy 85.1 kcal/mol, and the denaturation entropy 0.26 kcal/K∙mol.

Abstract Histoplasmosis is caused by the fungus Histoplasma capsulatum and is often fatal for individuals with acquired immunodeficiency syndrome (AIDS). Delayed diagnosis is a major factor in worsening coinfection, as it can be mistaken for other diseases. Thus, rapid identification of Histoplasma in immunocompromised patients is essential. Molecular techniques, particularly polymerase chain reaction (PCR), were used in this study to identify H. capsulatum in patients coinfected with histoplasmosis and AIDS. Blood samples from 14 individuals with AIDS and disseminated histoplasmosis were collected and analyzed. The PCR method successfully amplified the fungal region in whole blood samples, while PCR-RFLP analysis confirmed a consistent profile in the samples. Genetic sequencing further confirmed the fungal species. Compared to clinical tests such as fungal culture and urinary antigen detection, molecular analysis proved faster, more sensitive, and cost-effective. These molecular markers can potentially be incorporated into routine diagnostics in the future. Further studies are needed to expand and enhance this diagnostic approach, particularly in patients with nonprogressive clinical forms of histoplasmosis.

Abstract The diversity of endophytes and their ecological relationships with the endangered conifer Araucaria angustifolia (a critically endangered species) are unrevealed. This study aimed to characterize the diversity of endophytic fungi associated with A. angustifolia. To this end, we analyzed 90 fragments from five individuals collected from a mixed localized fragment in Guarapuava-PR, Brazil. The total DNA of 61 morphotypes was extracted and the Internal Transcribed Spacer (ITS) region was amplified and sequenced. The sequence analysis allowed the identification of 37 genera belonging to the phylum Ascomycota and the classes Eurotiomycetes, Dothideomycetes, and Sordariomycetes, divided into 11 orders and 13 families. Most of the isolated fungi belonged to the Sordariomycetes class (40%) and to the Xylaria genus (14%), while Eurotiomycetes was the minority class within the community. Our results reveal the high endophytic richness supporting the life cycle of A. angustifolia and reinforce the necessity for the conservation of this conifer, as many genetic resources can be lost owing to its irrational exploration.

Abstract The cultivation of marine shrimp is one of the fastest growing activities in the world. However, the emergence of diseases has resulted in a decrease in production and losses for the sector. Probiotics emerged as an option to the use of antibiotics to control these pathogens. The efficiency of applying this technology depends on the characteristics of the bacterial agents and their bioavailability in the shrimp intestine. The objective is to evaluate the viability and efficiency of bacteria isolated from the digestive tract of healthy crustaceans as probiotic agents in the cultivation of shrimp Litopenaeus vannamei. Eighteen strains of the genus Bacillus belonging to the following species were tested: Bacillus sp., B. cereus, B. thuringiensis, B. circulans, B. megaterium, B. subtilis and B. agaridevorans. Bacterial isolates were subjected to characterization as potential probiotics. The test results were considered satisfactory; thus, the tested strains have potential for use as probiotics in shrimp culture. Treatments that used of the genus Bacillus had reduced growth of the genus Vibrio after infection, both in the intestinal contents and in the intestine. With the results obtained, it can be suggested that further research be carried out on the probiotic potential of Bacillus sp.

Cloning, characterization and functional analysis of lichenase produced by Bacillus licheniformis RB16 isolated from cattle faeces EKIZ, DILEK OZGUN COMLEKCIOGLU, UGUR COMLEKCIOGLU, NAZAN AYGAN, ASHABIL

Abstract Lichenan, 1,3-1,4-β-Glucan, a linear polysaccharide exists in the cell walls of various cereals, has garnered attention for its industrial applications due to its enzymatic breakdown by lichenase enzymes. In this study, Bacillus licheniformis strain RB16, isolated from cattle faeces, was identified as a robust lichenase producer. The lichenase gene, licA, was successfully cloned and characterized. The cloned RB16 lichenase (LicA) demonstrated its highest activity level at pH 7.5. It also retained over 50% of its activity within the pH range of 6.0–8.5 but experienced a decline to 40% at pH 9.0. LicA was active at temperatures ranging from 25 to 65 °C with an optimum at 45 °C. LicA exhibited more than 60% of its activity at the temperature range of 35–55 °C. Zymogram analysis confirmed LicA’s lichenan-degrading ability and structural analysis revealed a stable enzyme structure primarily composed of random coils and extended strands. Although LicA exhibited low thermostability, consistent with its relatively low α-helix content, it demonstrated promising industrial potential. Evolutionary analysis placed LicA within a cluster of closely related Bacillus lichenases, particularly B. halotolerans, B. atrophaeus, and B. spizizenii. These findings expand our understanding of lichenases of Bacillus and underscore its potential for various industrial applications.

Abstract Hypostomus soniae is a small sized armored catfish endemic to the Tapajos River basin and ranked as one of the most exploited ornamental fish in the Santarem export marketplace. This study aims to evaluate distributional patterns of Hypostomus soniae and contribute to the species conservation in the face of development of the ornamental fish trade in the Amazon region. We compiled data associated with geographic coordinates in public repositories, supplemented with original field records. We compared our data to published records in the literature and museum collections to check for accuracy. To investigate the fishery and commercialization of H. soniae, we conducted interviews with ornamental fish stakeholders from the local trade. We also made direct observations in the fishing sites and export facilities in Santarem. A cluster analysis of the geolocation data was carried out to explore the spatial distribution patterns. The volume of captures and exportation of H. soniae decreased during the period 2020-2023. The occurrence of H. soniae was associated with annual rainfall ranging from 2,000 mm to 2,500 mm and concentrated in two municipalities of the State of Mato Grosso and two of the Para State. The species distribution area has been threatened, unfortunately, by fishermen who do not respect the laws that support artisanal fishing in the Amazon.

Abstract The Amazon floodplain is home to an extremely high diversity of fish, with lakes playing an important role in the establishment of this biological richness. These lacustrine environments are subject to constant fluctuations caused by the annual flood pulse, with local factors and other regional patterns also contributing to the variation in fish community structure. The present study verified how local (depth and transparency of the water, the size and species composition of the macrophyte stands) and regional factors (spatial distribution of the stands and the hydrological phase) influence the structure of the fish community of the floodplain lakes of the Môa River, in northern Brazil. Fish species richness was influenced by the depth of the water and the spatial distribution of the macrophyte stands. Fish species composition was influenced by local environmental variables, spatial structure, and the hydrological phase. However, variation partitioning indicated that only the hydrological phase explained the variation in fish composition. These findings indicate that the local environment, the spatial structure, and the hydrological phase drive changes in the structure of the fish communities associated with aquatic macrophytes in the floodplain lakes of the Amazon basin.

Abstract The agricultural sector is one of the most polluting economic activities, contributing significantly to greenhouse gas (GHGs) emissions. Brazil is one of the largest agricultural producers worldwide and plays a major role in reducing the environmental impact of this sector. Here, we aimed to determine the impact of the agricultural sector, with special attention to production, prices, and trade openness, on the short- and long-term GHGs emissions of Brazilian agriculture. Employing data from 1974 to 2019, we tested the cointegration of variables and compared the determinants of GHG emissions using Autoregressive Distributed Lag (ARDL) and Vector Error Correction Model (VECM) methods. Our results show a long-term equilibrium trend for Brazilian agricultural GHGs emissions, a result that correlates with emerging environmental compliance, and society demands the adoption of sustainable technologies, processes, and policies. In the short run, both cattle herds and Agricultural Added Value to GDP per capita showed an expected positive and significant contribution to GHGs emissions, while agricultural crop area demonstrated an inverse relationship. The trade openness index confirmed that foreign trade plays an important role in reducing GHGs emissions. The price index is not significant in our models. Both the private and public sectors have important roles in sustainable agriculture, especially in increasing system efficiency through the adoption of management and technologies that reduce GHG emissions levels.

Benthic foraminifera diversity from the south Atlantic Ocean: Tierra del Fuego and surrounding waters (South America) PÉREZ, SILVINA RAQUEL BERNASCONI, EMILIANA CANDEL, MARÍA S.

Abstract The present study provides a detailed record of foraminiferal fauna and their ecological implications from surface sediments from Atlantic shelf of Tierra del Fuego, Argentina. The foraminiferal assemblage is mostly composed by four main hyaline genera, such as Cibicidoides, Cibicides, Globocassidulina and Buccella, which allowed the identification of three environmental zones. Zone 1 (Z1, 37 to 90 m) encompasses the eastern Beagle Channel and San Sebastian Bay. The assemblage reflected well-oxygenated marine inner shelf habitat, adapted to cold temperate waters. Zone 2 (Z2, up to 98.4 m), is located around the southern tip of Tierra del Fuego. The assemblage suggested a deeper marine environment, well oxygenated and with higher energy, probably due to the effect of tides and mainly by the influence of Malvinas Current. Finally, Zone 3 (Z3, up to 195 m) is located furthest from the Atlantic coast and the assemblage suggested an environment characteristic of outer shelf, with well-oxygenated cold waters and high-energy environment, reflected by species adhered to the substrate and coarse sediments. The distribution and abundance of certain species showed the influence of the Malvinas Current, while others evidenced a contribution of the Cape Horn waters.

Abstract Under two detection schemes, this study analyzes one of the most destructive weather systems - the explosive cyclones - in the South Atlantic, from 2010 to 2020. Then, two methods are presented to study these systems: the Observational Method (OBSM) and the Automated Method (AUTM). The first uses visual analysis of the mean sea level pressure (mslp) fields and functions to identify the local minimums using the Grid Analysis and Display (GrADS) software. The second utilizes a function from OpenGrADS called mfhilo. It shows the local minimum in the grid using laplacian, magnitude, and percentile. Two shell algorithms for data manipulation are used for the AUTM: one to trace the cyclones’ trajectories according to a previously defined fixed area and the other to separate them into explosives. The OBSM methodology showed 271 cases averaging 25 yearly and revealed important characteristics regarding the intensities. According to AUTM’s methodology, from the 2705 ordinary cyclone cases identified, 299 are explosives. There is a clear seasonality pattern in the systems’ distribution along South America, similar to OBSM, but more highlighted. In summer, they concentrate at high latitudes, while in winter and spring, they are assembled near southern Brazilian and Uruguayan coasts.

Abstract The phylogenetic affinities of early dinosaurs are a controversial topic. There are disputing scenarios aiming to explain the evolutionary tree of these reptiles. A number of factors play a role on this issue. High levels of intraspecific variation experienced by the earliest forms are pointed by distinct authors as one of the main sources of phylogenetic biases. In the present study, we performed experimental analyses incorporating skeletally immature individuals as distinct operational taxonomic units in order to investigate the effect of ontogeny on the phylogeny and morphospace of early Late Triassic sauropodomorphs. The results support an “artificial” grouping of skeletally immature individuals of early dinosaurs in the phylogenetic trees, which is controlled by ontogenetic development. Such results are interpreted as the effect of ontogenetic control on the states of certain morphological characters, producing false synapomorphies. In addition, the morphological disparity analysis indicated a significant difference between the skeletally immature and mature individuals, corroborating previous assumptions. At this point, we suggest caution during the scoring process of early dinosaurs. Some approaches that avoid ontogenetically sensitive characters revealed a putative solution. Nevertheless, additional specimens and alternative tools are mandatory in order to properly deal with this issue.

Abstract The record of Mesozoic reptiles in the Northern region of Brazil is extremely limited, with the only definite occurrence consisting of two ziphodont teeth recovered from an oil well core in the municipality of Nova Olinda do Norte, Amazonas state, from strata of the Alter do Chão Formation (Cenomanian) of the Amazonas Basin. In this study, we aim to reevaluate MCT.R.514 using the most recent methodologies available for the identification of isolated theropod teeth. The cladistic analyses recovered MCT.R.514 as a possible metriacanthosaurid or non carcharodontosaurid allosauroid while the LDAs showed affinities between the specimen and Abelisauridae, Piatniktzysauridae, and Tyrannosauridae. MCT.R.514 was assigned as an abelisaurid due to its overall morphology that lacked metriacanthosaurid synapomorphies (i.e. spiraling mesial carina, strongly labially displaced distal carina, and well-marked mesiolingual longitudinal groove), while maintaining homoplastic features between both groups (i.e. irregular enamel texture, a lingually biconcave cross section, and a straight distal margin). This, together with a mostly Laurasian distribution of Metriacanthosauridae favors the assignment of MCT.R.514 as an abelisaurid as the most parsimonious hypothesis in this occasion. The identification of the specimen as an abelisaurid further expands the still scarce “Mid’’ Cretaceous record of this clade in Brazil.

Abstract Pacu (Piaractus mesopotamicus) is a fish with a high production potential in Brazil. However, one limitation is the excessive amount of ether extract in its carcass, an undesirable characteristic for the consumer. One approach to overcome this limitation is to improve carcass quality through zootechnical additives such as caffeine. The aim of this study was to evaluate the effect of supplementing the diet of pacu with caffeine on cut yield, biological indices, and carcass composition. Two hundred pacu with an initial weight of 1,687 g were used. The animals were allocated to 20 aquaculture cages of 1 m³, with 10 animals per cage. A completely randomized design with four treatments and five replicates was used. The treatments evaluated consisted of four inclusion levels of caffeine: T1 = 0.00 g; T2 = 0.16 g; T3 = 0.32 g, and T4 = 0.48 g caffeine.kg-1 of feed. The findings show that caffeine can be recommended as a diet supplement for carcass improvement of pacu, reducing the fat content and increasing the protein content of the carcass. Caffeine up to 0.32 g.kg-1 of feed can be added to the diet of pacu without affecting its performance or cut yield.

Abstract This study was conducted to determine the best combination of protein sources in diets for jundiá, based on growth, metabolism, and nutrient deposition. Five protein combinations were tested: casein + fish meal (control), casein + gelatin, casein + albumin, casein + albumin + fish meal, and albumin + fish meal, in diets containing 370 g Kg-1 of crude protein and 13.4 MJ Kg-1 of digestible energy. The fish (9.38 ± 0.12 g) were allocated in a water recirculation system at a density of 3.35 g L-1 per experimental unit and fed until apparent satiety for 40 days with the diets. The fish fed with the control diet had the highest final weight, specific growth rate, protein and feed efficiency ratio, protein retention, and best apparent feed conversion. On the other hand, fish that received casein + albumin and albumin + fish meal diets showed worse results in growth and body protein retention, low trypsin and chymotrypsin activity, and high intestinal amylase activity. Therefore, the combination referred to as control (casein + fish meal) conclusively provides the best rhythm for nutrient digestion and metabolism processes, enabling fish to reach greater growth and retention of body protein with low whole-fish fat content.

Abstract Many aspects of the ecology, evolution and social behavior of wild-living primates remain un-explored and require further investigation. While long-term field studies are crucial for addressing conservation concerns for many primates’ species, acquiring the necessary data is often challenging, often due to difficulties in locating study groups. Radio-telemetry has significantly facilitated the study of primates and other animals living in tropical forests. However, there are important practical challenges in the process of capturing and releasing animals after placement of telemetry collars. In this study, we report guidelines and considerations for capturing and collaring wild capuchin monkeys, Sapajus nigritus cucullatus, in the Atlantic Forest of Argentina. Our ultimate goal is to contribute to making captures safer, preventing harm and stress to animals when using radio-telemetry in monitoring strategies for conservation of this primate species. These methods can be useful for researchers using field capture and radio-telemetry for monitoring groups or populations of wild primates, specifically wild Sapajus.

Abstract This study investigated the spatial distribution, parasite interactions, and affinities of Branchiura parasites according to macrohabitat and surface type on fish from the Araguari River, Amapá State, in the Brazilian Amazon. This study presents novel information on the interactions of branchiuran species with host fish, and how the morphology of their attachment structures is associated with different host surface morphologies, with and without scales. A total of 558 fish specimens of 26 species were collected, of which 109 fish (10 species) were parasitized by at least one branchiuran parasite; these were selected for further study. The parasites were collected from the buccal cavity, gills, fins, and integument of the host fish. Parasites with suction discs (Argulus) were mainly associated to hosts with small scales and no scales, while parasites with hooks (Dolops) were mainly associated to hosts with large scales. Overall, parasites with suction discs were more specialized per site than those with hooks.

Abstract Fish parasites are an important part of aquatic biodiversity and knowing these species and their interactions with their hosts helps in monitoring the aquatic biota. The present study investigated the ectoparasite crustacean fauna of ten fish species from the upper Araguari River, in the state of Amapá, northern Brazil. A total of 508 fish were collected and analyzed from July to November 2014, of which 82.6% (109) were parasitized by one or more crustacean ectoparasite species. In the ten host fish species, a total of 308 ectoparasite specimens were collected, from 12 taxa, such as Argulus multicolor Stekhoven, 1937, Argulus spinulosus Silva, 1980, Argulus sp.1, Argulus sp.2, Argulus sp.3, Dipteropeltis sp., Dipteropeltis hirundo Calman, 1912, Dolops bidentata Bouvier, 1899, Dolops striata Bouvier, 1899 (Argulidae), Braga fluviatilis Richardson, 1911, Braga amapaensis Thatcher, 1996 (Cymothoidae) and Excorallana berbicensis Boone, 1918 (Corallanidae). Higher levels of prevalence and abundance were recorded for Hoplias aimara (Valenciennes, 1847) and Tometes trilobatus Valenciennes, 1850, respectively. These ectoparasites were found in the fins, integument, mouth, and anus of the host fish. Argulus sp.2 and D. bidentata were the most abundant parasites (65.1%), and had the highest species richness. This study registered 36 novel host-parasite interactions, and thus represents a new record for all host species here examined.

Abstract The current study aims at using non-hatchable artemia eggs of local origin and making use of these eggs by decapsulating and presenting them as food for the larvae of the Cyprinus carpio as a source of animal protein with high nutritional value instead of throwing them away. The results showed that the second parameter (A2) was highly significant at the level (P≤0.05) in the growth rates of the larvae that were fed on decapsulated artemia eggs alone, and it was better than the two control parameters (A1), in which the larvae were fed with feed designated for Cyprinus carpio fish. It also outperformed the third parameter (A3), in which the feed was mixed with artemia eggs with 50% decapsulation, which also outperformed the control parameter with high significance at the same level (P≤0.05).

A novel approach to urban turtle sampling: Assessing Hookless Fishing with clip and two conventional methods ROCHA, SABINE B. GROU, CARLOS EDUARDO V. ROUCO, CARLOS

Abstract To address urban turtle sampling challenges, we presented Hookless fishing with clip, a cost-effective method for sampling this important group. Effectiveness, biases and potential advantages were analysed in comparison to two commonly used methods (funnel trap and hand capture). Fieldwork was conducted between August and November/2021 in four areas in Brazil, using the three methods simultaneously. A total of 195 turtles from four species (Phrynops geoffroanus, Hydromedusa tectifera, Trachemys dorbigni and T. scripta elegans) were captured. Funnel trap demonstrate a significantly higher capture than hand capture, while Hookless fishing showed no significant difference in captures compared to funnel trap. The highest catch per unit effort values were observed for the new method (0.37) and the funnel trap (0.34). Despite being widely used, funnel traps were the only method to exhibit male bias. Our findings revealed that Hookless fishing with clip exhibited remarkable capture efficiency, cost-effectiveness, and ease of transport and utilization; however, it requires operator presence. Nevertheless, the proposed method, both as the primary or auxiliary approach, appears efficient in enhancing captures and reducing costs and risks. This innovative method has the potential to assist researchers studying omnivorous and carnivores freshwater turtles in environments worldwide, especially in human settlements.

Abstract We studied the relationship between shape, size, and developmental time in the embryonic ontogeny of 15 species of the frog genus Physalaemus. As in other anuran exotrophic embryos, shape changes are correlated with size increase and mainly concern tail elongation, decrease in body height, and increase in fin height. Size ranges and developmental times vary interspecifically. Embryos of the P. signifer Clade and the P. gracilis Group are among the largest, are slightly peramorphic, and develop fast regarding congeneric species. Embryos of P. cicada combine the smallest sizes with fast development and the most peramorphic shapes. The paedomorphic shapes of embryos of P. biligonigerus and P. henselii groups are correlated with fast vs. slow developmental times respectively. Trajectories in the P. cuvieri Group are diverse and in general differ in size and developmental time. The embryos of P. cristinae and from the Argentinean lineage of P. cuvieri stand out with the longest development. Sequences of developmental events are overall conserved in the genus, and main differences concern mouthpart ontogeny. This study constitutes the first attempt to evaluate morphological, allometric, and heterochronic parameters of the early ontogeny of anurans and how these can vary and contribute to diversification in taxonomic groups.

Abstract Concentrated supplementation of ewes is a strategy to increase productivity. The objective was to evaluate the effects of supplementation in the diet of ewes before, during and in the final third of pregnancy and lactation on, the performance and production and composition of colostrum and milk. Forty animals were distributed in a completely randomized design, into the following treatments: CONT = control treatment with mineral salt supplementation only, SSREPRO = supplementation started 20 days before the estrus synchronization (ES) protocol, SSPREG = suplementação a partir da confirmação da gestação (60 days after ES), and SEPREG = supplementation in the third end of pregnancy (90 days after ES). Supplementation adoption changed (P < 0.05) the intake of organic matter, crude protein, neutral detergent fiber, the percentage of total digestible nutrients, and digestibility of dry matter, acid detergent fiber, non-fiber carbohydrates, and ether extract with their respective intake, in addition to colostrum and milk production and composition and animal performance. No difference (P > 0.05) was observed for organic matter, crude protein, and neutral detergent fiber digestibility and total digestible nutrient intake. Thus, supplementation in the final third of pregnancy may result in heavier lambs at weaning, reduction in slaughter time.

Abstract In the Neotropical Region, Phlugidini is the most diverse tribe of Meconematinae, with 62 valid species in 10 genera, six of them recorded from Brazil. Brachyphlugis gen. nov. is described herein based on two new species, Brachyphlugis longicercalis sp. nov. (type species) and Brachyphlugis curvata sp. nov., collected in Parque Nacional do Itatiaia and Reserva Biológica do Tinguá, both in the Atlantic Forest of Rio de Janeiro State. The new genus is most similar to Neophlugis, but it can be distinguished from the latter and other neotropical Phlugidini by the following combination of characteristics: (1) mandibles symmetrical; (2) tegmen extending beyond metanotum; (3) pronotal disk posterior margin convex; (4) tenth tergite strongly projected and downcurved; (5) male cercus pronounced with a larger base and acuminate apex; (6) paraproct modified with developed spine; (7) male subgenital plate with two distolateral projections connected to a pair of styles; and (8) female subgenital plate wider than long, posterior margin with two projections. An up-to-date key to neotropical Phlugidini genera is also provided.

Abstract Sapajus nigritus (Primates: Cebidae) is endemic of the Atlantic Forest, occurring from the Southeastern and Southern regions of Brazil to Northeastern Argentina. This species plays a role in two ecosystem services: seed dispersal through endozoochory and insect population control in agricultural plantations and forest environments. Fruits and invertebrates represent approximately 90% of their diet, and there is a large number of insects in the diet of S. nigritus in urban and conserved areas. However, it is known that insect diversity decreases in anthropized environments. Our objective was to identify the insects present in the fecal samples of S. nigritus from five Conservation Units in Rio de Janeiro state, Brazil. We aim to estimate the percentage of each taxon of insects found in feces either, hypothesizing that there are a greater variety of insect species in the diet of S. nigritus that inhabit preserved forested areas. A fecal screening was conducted using a light microscopy and the insects were identified based on their external morphology. Insect fragments were found in eight out of ten fecal samples of S. nigritus, revealing that they belonged to insects from five orders: Hymenoptera, Hemiptera, Orthoptera, Coleoptera and Blattodea, suggesting a good conservation status of the sampling areas.

Abstract Several countries and non-governmental organizations have discussed the use of animals in industry and biomedical areas. This work shows the progression of animal’ rights for scientific purposes in Brazil and how Brazilian Councils have advanced to follow worldwide regulations. Since the first rules about animals’ usage in Ireland in 1635, the British Cruelty to Animals Act in 1876, and the Brazilian animal protection rules in 1924 and 1934, most worldwide actions culminated in the Universal Declaration of Animal Rights (1978). In 1979, the Brazilian Law 6.638 displayed directives for didactic-scientific practice of vivisection. In 2008, the Arouca Law 11.794 filled regulatory gaps and created the National Council for the Control of Animal Experimentation (CONCEA). In 2014, the CONCEA incorporated the 3R’s philosophy and recognized substitute techniques, but only in 2023 it prohibited vertebrate animals in scientific research, development and control of personal hygiene products, cosmetics and perfumes. It is clear current Brazilian and international rules are unable to cover all aspects of animal wellbeing, even for regulations of commercial issues. Certainly, innovative tools, as organ-on-chip, in vitro techniques and bioinformatical advancements will provide a crucial animal welfare and new laws will minimize animal pain and distress, including for disregarded invertebrates.

Abstract This study examined the effects of wheat and corn gluten added to lamb diets as a unilateral protein source on some microbial and chemical properties of Musculus Longismus dorsi (LD), determination of intramuscular and tail fat profile. It was found that TBARS levels in LD muscle on the days of storage were highest in the wheat gluten-treated groups (p<0.01). It was found that the changes in pH values in LD muscle were different on days (p<0.05). It was found that the change of L*, a ve b values on days differed between groups during the storage period (p<0.05). It was found that the numbers of TMAB, Enterobacteriaceae, Lactobacillus spp., Pseudomonas spp. and TPAB changed significantly (p<0.05) during the storage process. While a significant difference was found between the MUFA levels of dorsal muscle intramuscular adipose tissue of the groups (p<0.05). As a result, it was determined that the metabolic differences of the one-way protein sources fed to the lambs in the digestive system and other organs had an effect on the meat quality, intramuscular fat and fatty acid profile of the tail.

Abstract Ranacephala hogei is a South American freshwater turtle considered one of the 25 most endangered chelonian species in the world. Endemic to the Atlantic Forest hotspot of southeastern Brazil, the conservation status of R. hogei is subject to continuous assessment at various levels. However, the scarcity of data regarding this species, particularly its geographic range, challenges these evaluations. In an effort to address these gaps, we conducted a comprehensive long-term inventory using different methods to study this species. Our efforts resulted in a 144% increase in documented occurrence points, including a new hydrographic basin and protected areas. By combining historical and current records, we have observed the persistence of R. hogei in rural areas, even in locations where its extinction was previously predicted. Consequently, our data significantly contribute to supporting future conservation assessments.

Abstract Recent research has demonstrated the increasing interest in using insects for the extraction of bioactive compounds, particularly peptides. These compounds offer a spectrum of beneficial physiological effects. The aim of this study was to standardize a methodology for obtaining bioactive peptides from Tenebrio molitor and evaluate its physicochemical characterization, antioxidant, and antimicrobial potential. Six assays were carried out to hydrolyse larvae protein, with variations in Alcalase concentration (0.04 to 0.08%) and reaction time (3 to 8 h). The results indicated that the process applied to defatted mealworm flour was effective in reducing lipids by 82.5%. Consequently, it was an observed increase of 38.4% in protein content. Additionally, an increase in glycogen content was found in defatted mealworm flour (177 µmol glucose g-1 sample) and peptides (152.81 µmol glucose g-1 sample). The degree of hydrolysis was higher in assays with longer hydrolysis durations (8.14 - 8.38%). The antioxidant capacity was 12 to 14% lower in assays with an incubation time of 8h. In this sense, the methodology proposed in the present study proved to be efficient in obtaining bioactive peptides from T. molitor.

Taxonomic study of Hymenochaetaceae species (Agaricomycetes, Basidiomycota) in the municipalities of Tomé-Açu and Bujaru, Pará, Brazil FREIRE, RICHARD BRUNO M. SOARES, ADRIENE MAYRA S.

Abstract The family Hymenochaetaceae includes a diversity of 893 species described around the world. Its representatives are known by their usually rusty colored basidiomes with a poroid hymenial surface, hydnoid or smooth, woody consistency, and wide morphological variation regarding the arrangement on the substrate. They behave as saprophytic, parasitic, ectomycorrhizal and play a fundamental role in the decomposition of wood in forest ecosystems. In the Brazilian Amazonia region, approximately 40 species of Hymenochaetaceae are currently recorded. The main goal of this study was to increase the knowledge on Hymenochaetaceae from the Brazilian Amazonia. Collections were carried out between October 2021 and April 2022 in the state of Pará, municipalities of Tomé-Açu and Bujaru, to expanding the knowledge of this fungal family to the Brazilian Amazonia. A total of 15 specimens were identified, distributed in seven genera and 12 species. Four species are new records for the state of Pará (Fomitiporia apiahyna, Phellinus neocallimorphus, Phellinus sancti-georgii, and Sclerotus extensus) and two of them are new records for the Brazilian Amazonia (P. neocallimorphus and P. sancti-georgii). Our findings contribute to taxonomic knowledge of this family in the Brazilian Amazonia and reduce the information gaps about the diversity of species.

Abstract The present study modeled the adsorption process of the drug diclofenac sodium on activated charcoal. For this purpose, a mass balance-based model was used considering a fixed bed column. The mass transfer rate in the solid phase was represented by a driving force model proposed in this study, and a gamma exponent with a range of 0>γ≤2 was assigned to the model. Different isotherms were adopted to represent the equilibrium at the solid/liquid interface: the Langmuir, Freundlich, Sips and Redlich-Peterson isotherms. Abstract With the evolution of energy storage, Thermal Runaway (TR) stands out as the most critical safety concern for Lithium-Ion Batteries (LIBs). This study employs a prismatic lithium battery with a nominal capacity of 40Ah, featuring Li(Ni0.6Co0.2Mn0.2)O2 as the cathode material. The investigation delves into the thermal runaway characteristics of the battery at 25%, 50%, 75%, and 100% State of Charge (SOC) in a nitrogen environment. The findings indicate: 1) an ascending trend in the highest temperatures at various points within the battery as SOC increases, accompanied by a declining trend in normalized gas production and a non-linear relationship between the heat released during TR and the stored electrochemical energy; 2) the highest temperature point within the battery consistently resides at the surface, offering insights for the temperature monitoring of the Battery Thermal Management System (BTMS); 3) a direct correlation between higher SOC and increased material ejection, with a mass loss rate of 25.8% at 100% SOC, a static total gas production of 2.45 mol, and a maximum explosion index of 0.2886 kPa⋅m⋅s⁻¹.

Abstract As it flows through the city of São Paulo, the Tietê River receives heavy discharges of industrial effluents and domestic sewage, resulting from the city’s continuous urban expansion and the inadequacy of its sanitary sewage system. This study focused on an analysis of the efficiency of PGα21Ca and quaternary ammonium tannate, water purification products, based on coagulation-flocculation and sedimentation tests, followed by treatment with a hydrodynamic cavitation reactor associated with ozonation in the treatment of Tietê River water. The removal of turbidity, apparent color, and chemical oxygen demand (COD) were evaluated. Jar testing assays were conducted, and the best turbidity removal rates were obtained with a concentration of 300 mg L-1 for PGα21Ca and 150 mg L-1 for quaternary ammonium tannate. The coagulation-flocculation treatment removed approximately 93% of turbidity for both coagulants. After combining coagulation-flocculation with hydrodynamic cavitation with ozonation, the final COD removal rate applying PGα21Ca was 47.63% in 1 hour of reaction, while that of quaternary ammonium tannate was 40.13% in 2 hours of reaction. Although the results appear to indicate the superior performance of PGα21Ca, it should be noted that the treatment with quaternary ammonium tannate also provided good results in reducing turbidity, COD, and apparent color, using a smaller dose of this coagulant and that its use may be more advantageous from an environmental point of view, due to its natural composition.

Abstract This study aimed to produce water-soluble oat extract enriched with mango peel flour (MPF) as a source of active compounds and to use this ingredient as a partial substitute for whole milk in Greek yogurt (GY) for its nutritional enrichment. Enriched water-soluble oat extracts (EWSOE) were produced with different concentrations of MPF (0%, 1%, 1.5% and 2%) and characterized in relation to pH, titratable acidity, soluble proteins and total phenolics. Three GY formulations were prepared by partially replacing whole milk with EWSOE and the best formulation (in relation to sensory analyzes and phenolics compounds) was selected for storage study, chemical characterization, and sensory acceptance testing. MPF addition increased soluble proteins and total phenolics in EWSOE. GY formulations prepared with EWSOE had similar sensory scores. During storage, GY prepared with EWSOE containing 2% MPF exhibited changes in pH and titratable acidity and a reduction in total phenolics. Color parameters, cholesterol, and fatty acid composition did not change over 21 days of storage. The major fatty acids in GY were oleic and palmitic acids. The selected product had low lactose content (1.2%), achieved satisfactory sensory acceptance in relation to the evaluated attributes, and had lipid (~6.19%) and protein (~3.96%) contents within regulatory requirements. Additionally, EWSOE is a valuable ingredient in GY preparation, offering beneficial nutritional and functional properties.

Abstract The high performance of biomass and metabolite biosynthesis by photosynthetic microorganisms is directly influenced by the cultivation system employed. Photobioreactors (PBRs) stand out as controlled and fundamental systems for increasing the production of biocompounds. However, the high costs associated with these systems hinder their viability. Thus, a more practical and economical approach is necessary. Accordingly, this study aimed to design and evaluate low-cost flat-panel photobioreactors on a laboratory scale for the cultivation of photosynthetic microorganisms, using economical materials and instruments. Additionally, internal optimization of the low-cost system was aimed to maximize growth and biomass production. The PBRs were designed and built with uniform dimensions, employing 4 mm translucent glass and agitation through compressors. The internally optimized system (PBR-OII) was equipped with perforated acrylic plates used as static mixers. To evaluate the performance of the low-cost PBR-OII, a comparison was made with the control photobioreactor (PBR-CI), of the same geometry but without internal optimization, using a culture of Synechocystis sp. CACIAM 05 culture. The results showed that the PBR-OII achieved maximum biomass yield and productivity of 6.82 mg/mL and 250 mg/L/day, respectively, values superior to the PBR-CI (1.87 mg/mL and 62 mg/L/day). Additionally, the chlorophyll concentration in the PBR-OII system was 28.89 ± 3.44 µg/mL, while in the control system, the maximum reached was 23.12 ± 1.85 µg/mL. Therefore, low-cost photobioreactors have demonstrated to be an essential tool for significantly increasing biomass production, supporting research, and reducing costs associated with the process, enabling their implementation on a laboratory scale.

Abstract This study addresses the prediction of fatigue life in SAE AMS 7475-T7351 aluminum alloys under variable loads, commonly used in the construction of aircraft fuselages. The main objective of the research was to develop a numerical-experimental procedure to analyze crack growth, using the Walker’s approach which considers the effects of the stress ratio R on the fatigue crack growth rate da/dN, combined with the Finite Element Method and Linear Regression of the Stress Intensity Factor. Observations showed that Walker’s model effectively consolidated fatigue crack propagation data for various stress ratios when applied longitudinally to L-T rolling orientation, due to low dependence of exponent m on R-value in da/dN equation. Simple averaging of m values effectively calculated Walker’s exponent. The methodology employed experimental tests following ASTM standards for tension, fracture toughness, and fatigue, complemented by Finite Element Method (FEM) simulations. The Walker’s model proved more effective, while the Paris-Erdogan model, which ignores the R effect, resulted in overly conservative service life estimates. The principle of similitude suggests that this methodology could be effective in predicting fatigue life in cases with complex geometries, where calculating the Stress Intensity Factor Fracture parameter is challenging and the Finite Element Method shows efficiency.

Abstract Upcoming Earth Exploration Satellite Services (EESS) missions, especially to monitor Brazilian diversified biomes, will require progressively higher data rates for downlink transmissions, besides the ability to share its frequency spectrum with cellular base stations. Both impact issues on spectral efficiency (in bps/Hz) and coexistence in frequency, time, location, etc. This paper proposes a technique suitable for LEO Earth Observation Satellites (EOS) by combining two strategies. We initially present the Cognitive Radio (CR) spectrum awareness and exploitation approaches to propose techniques for improving their uses. Next, we detail the Adaptive MODulation and CODing (MODCOD) techniques (ACM) based on DVB-S2X systems to increase RF power and spectral efficiencies. Finally, we evaluate our solution by monitoring the Signal to Interference plus Noise Ratio (SINR) and combining CR/MODCOD techniques. Two case studies are presented that demonstrate the proposed approach on Brazilian satellites developed by the National Institute for Space Research (INPE). A real in-situ characterization of the interfering scenarios was performed during the passes of the two EESS satellites that proves the effectiveness of spectral efficiency and coexistence.

Abstract Biorefineries require low-cost production processes, low waste generation and equipment that can be used not only for a single process, but for the manufacture of several products. In this context, in this research a continuous 3D printing microbioreactor coupled to an Arduino-controlled automatic feeding system was developed for the intensification of the ethanol production process from xylose/xylulose (3:1), using a new biocatalyst containing the co-culture of Scheffersomyces stipitis and Saccharomyces cerevisiae (50/50). Initially, batch fermentations of monocultures of S. cerevisiae and S. stipitis and co-culture were carried out. Subsequently, the immobilized co-culture was used as a biocatalyst in continuous fermentations using the developed microreactor. Fermentations carried out in the microbioreactor presented a 2-fold increase in the ethanol concentration and a 3-fold increase in productivity when compared to monocultures. The microbioreactor developed proved to be efficient and can be extended for other bioproducts production. This approach proved to be a promising alternative for the use of the hemicellulose fraction of biomasses without the need to use modified strains.

Abstract Echoing Arturo Casadevall and Ferric Fang in their Reforming Science: Methodological and Cultural Reforms, “great human enterprises must undergo periodic cycles of self-examination and renewal to maintain their vigor”. Especially in the last decade, the research culture has undergone such cycles, partially driven by countercultural transformations that have been reshaping assumptions towards reward-deserving achievements. Addressing retractions is among the challenges in this culture. This work builds upon research carried out at the University of California, San Diego (UCSD), which explored the views of 224 reviewers serving on panels for the US National Science Foundation, the National Institutes of Health, among others. We show results of a survey that add to our previous data. It was sent to a population of 1,089 corresponding authors affiliated with institutions from the 20 most productive countries in biomedical fields. We explored how corresponding authors of at least one retracted publication issued between 2013 and 2015 in biomedical journals envisioned the impact of different types of retractions on the careers of the first and corresponding authors. As such impact (if any) is not always immediate, we selected this time frame to ensure that potential respondents would have tangible post-retraction experience.
